Preformulation: Important tool in drug developement
Submitted by Shiv Majumdar on Sat, 09/05/2009 - 10:27Preformulation:
• a stage of development during which the physicochemical properties of drug substance are characterized.
When?
• If the drug shows sufficient activity in animals & is to be evaluated in humans.
Focus…
• On physicochemical properties of a new compound which may affect the drug performance & development of efficacious dosage form.

Dissolution Test : Limitations
Submitted by Shiv Majumdar on Fri, 09/04/2009 - 11:29We all studied dissolution test as the important thing that mimic the in-vivo fate of drug/dosage form.
Despite the fact that dissolution testing is a fundamental part of drug product development and regulatory approval process, it has been a subject of frequent criticism for its limitations of sometimes being non-discriminating, over-discriminating, and formulation specific IVIVC.

Concept of mapping in IVIVC
Submitted by Shiv Majumdar on Wed, 09/02/2009 - 05:43Mapping is a process which relates Critical Manufacturing Variables (CMV), including formulation, processes, and equipment variables that can significantly affect drug release from the product, to a response surface derived from an in vitro dissolution curve and an in vivo bioavailability data.

POROUS TABLET: Novelity in Tablets
Submitted by Shiv Majumdar on Tue, 09/01/2009 - 04:12A volatile solid (e.g Urethrane or ammonium bicarbonate) is added to a standard, directly compressible formulation.
After the tablet compression, the volatilizable solid is removed by a freeze-drying or heating process.
Water easily penetrates through the pores & promotes rapid disintegration.
e.g. Lyoc and Zydis Tablets

HIESTAND TABLETING INDICES: Important Parameter to be considered during formulation
Submitted by Shiv Majumdar on Mon, 08/31/2009 - 12:09• Hiestand quantify rationally the compaction properties of pharmaceutical powders.
• The results of this work are three indices known as Hiestand Tableting Indices.
– The Strain Index (SI)
– The Bonding Index (BI)
– The Brittle Fracture Index (BFI).
• The SI is a measure of the internal entropy, or strain, associated with a given material when compacted.
