Copyright © 2003 American Chemical Society The oxidation of an -hydroxy acid to a keto acid is catalyzed by a number of flavoenzymes, including flavocytochrome b2, glycolate oxidase, lactate oxidase, lactate monooxygenase, mandelate dehydrogenase, and -hydroxy acid oxidase. The three-dimensional structures of mandelate dehydrogenase, glycolate oxidase, and flavocytochrome b2 show that these are homologous proteins with TIM barrel structures. The sequences of the other enzymes and the conservation of critical active site residues are consistent with all members of this family having a similar structure and presumably a common catalytic mechanism for -hydroxy acid oxidation.
